目的观察嗜酸乳杆菌对溃疡性结肠炎(UC)小鼠结肠黏膜黏液细胞、黏蛋白MUC2的表达,了解其治疗UC的效果及可能作用机制。方法60只BALB/c小鼠随机分为健康对照组、嗜酸乳杆菌(高、低剂量)组、阳性对照(美沙拉嗪)组、阴性对照(冻干赋形剂)组、模型对照组。健康对照组自由饮水,其余各组自由饮用3%葡聚糖硫酸钠7d造模,同时给予嗜酸乳杆菌、美沙拉嗪灌胃治疗。黏液组织化(A学B/PAS)法计数结肠黏液细胞;免疫组织化学检测黏蛋白MUC2的表达抵;采用反转录聚合酶链反应(PCR)检测MUC2的基因表达。结果模型对照组、阴性对照组与健康对照组相比小鼠结肠黏液细胞数、黏蛋M白UC2含量减少,MUC2的基因表达下降;嗜酸乳杆菌和美沙拉嗪不同程度增加结肠黏液细胞数量、黏蛋白含量,促进MUC2的基因表达,嗜酸乳杆菌高剂量组效果较显著。结论嗜酸乳杆菌对急性期UC小鼠有治疗作用,疗效与剂量呈正相关;其机制可能与嗜酸乳杆菌促使结肠黏膜MUC2的表达上调和结肠黏膜中黏液细胞数量增多有关。
